Abstract

Nitric oxide (NO) radical species is involved in many biological processes. Notwithstanding the great importance of NO in biological systems, studies on their pharmacological and physiological properties are limited because of their high reactivity and short half-life time. Thus, the development of compounds that can release NO, can be an alternative to solve these problems and potentially be used as therapeutic agents. One of these possibilities is the anticancer action of NO. Based on that this project has the general objective to synthesize species that can act as donors agents of NO by reduction process and evaluate the release site of NO through the use of luminescent compounds. In these studies are summarized species [Ru (bpy-R)2(BODIPY)NO](PF6)3 (I) wherein R = bpy ligand-fluorophore (BODPY); [Ru(bpy-R)2(bodipy)NO]3+ (bpy-R = 2,2'-bipyridine). The chemical and kinetic characterization of these systems will be evaluated by spectrophotometric and electrochemical techniques. Nitric oxide release kinetics from the reduction of [Ru (bpy-R)2(BODIPY)NO]3+ will be evaluated using nitric oxide sensor and spectroscopic methods. Cytotoxicity will be determined by the MTT assay and flow cytometry. The comparison of the cytotoxic effect of all the species [Ru (bpy-R)2(bodipy)NO]3+ will evaluate the "uptake" of the compound due to the structural change of the bipyridine ligand. The mitochondrial membrane potential is estimated using Rhodamine 123. (AU)
