Obesity is the most common metabolic disorder in the world, characterized by excess adipose tissue (AT). The literature thus presents several strategic approaches to deal with obese individuals, including food restriction and physical exercise. Thyroid hormones (TH), principally triiodothyronine (T3), they are fundamental to proliferation control and differentiation of AT. The irisin is a protein synthesized and secreted by skeletal muscle of humans and rats is produced after physical exercise and acting on TA stimulating UCP-1, thus leading to release temperature and thereby decrease the fat. Both T3 as irisin can initiate actions resulting in thermogenesis and energy consumption. This study with use of the human cell culture adipocytes aims to identify, by analyzing the transcriptome based on RNAseq, the genes up or down regulated by the action of T3 and/or irisin. The action of T3 on irisin present in TA will be measured by Western blot (WB). The presence and amount of UCP-1 will be evaluate by WB. The DNA damage will be assessed by quantifying the hydroxydeoxyguanosine-8 by ELISA, besides the evaluation of the difference amount of lipids within cells after the treatments. These results this research provide a comprehensive view of the action of triiodothyronine and irisin in the expression of numerous genes in the TA, thus opening the way to explain the pathophysiology of the disease.
