This project aims at studying the action of carbon nanotubes and algal polysaccharides as mediators for the immobilization of enzymes as Langmuir and Langmuir-Blodgett (LB) films constituted of lipids, which serve as bioinspired systems. Lipids of various kinds will be used, among the candidates are the dipalmitoyl phosphatidylcholine (DPPC), dipalmitoylphosphatidylglycerol (DPPG) and dioctadecyl dimethylammonium (DODAB), which will be co-spread on the air-water interface with the enzyme urease. The incorporation of carbon nanotubes and algal polysaccharides, or via the aqueous subphase or via immersion of pre-prepared film in the solution / dispersion will be evaluated in order to improve the optical and electric signals in the detection of the enzymatic activity. The incorporation of the enzyme in the air-water interface in the mixed lipid system / polysaccharide / nanotube is measured by surface pressure and surface potential measurements, kinetic curves adsorption, compression isotherms, microscopy Brewster angle (BAM,) and polarization-modulation reflection-absorption infrared spectroscopy (PM-IRRAS). The films will be transferred to solid supports using the Langmuir-Blodgett technique and characterized by spectroscopic techniques. The results will be evaluated from the perspective of possible implications in nanobiotechnology, inferred from the effects caused by carbon nanotubes and algal polysaccharides in artificial membranes, especially as the incorporation of enzymes.
