Abstract

Introduction: Evidence suggests the existence of daily variations of the control of the hypercapnic chemoreflex, and has been assigned to orexinergic neurons in the Lateral Hypothalamus and Perifornicial Area (LH / PFA) the role of mediator of the circadian variations in central chemosensitive function, but the mechanisms involved are not well known. Recent results from our group suggest that metabotropic glutamate receptors in the LH/PFA perform an inhibitory modulation of hypercapnic ventilatory response during wakefulness but not during sleep, but it is not known the subtype of metabotropic receptors involved in this effect. Objective: The main purpose of this project is to investigate the possible role of glutamatergic neurotransmission via metabotropic receptors of group II in the LH/PFA in the modulation of ventilatory response to hypercapnia in rats during wakefulness and sleep, in the dark and the light period of the light/dark cycle. Methods: To this end, unilateral injections of LY 341495 (glutamatergic metabotropic antagonist group II) will be performed into the LH/PFA of male Wistar rats, and the pulmonary ventilation will be evaluated in a plethysmography system. It will be studied the effects of this approach in the ventilation in air and 7% CO2, during sleep and wakefulness, in both the light and the dark period. (AU)
