Abstract

Prostate cancer (PCa) is the most common malignancy in men and the second leading cause of male cancer-related deaths in the Western world. Studies emphasize the role of stroma compartiment on initiation and progression of CaP. In an ending project, two knockout mice models, the first one exhibits prostate-specific homozygotic deletion of Pten and the other exhibits prostate-specific homozygotic deletion of p53 and pRb, were used to produce trasncriptomes, by next generation sequencing, of normal ventral, lateral, dorsal and anterior prostatic lobes and tumoral samples at different stages of development and progression. Previous analysis of these transcriptome have reveled alteration in the expression of reactive stroma related-genes, such as increased expression of MMPs, type I and type III collagen alpha chains, tenacin C and in the genes of Syndecan 1-4 heparan sulphate proteoglycans family and their cytoplasmatic binding protein Syntenin 1. The aim of this project is to characterize by immunohistochemistry the tissue localization of the 4 members of Syndecan family proteoglycans and Syntenin 1 in the normal and tumoral prostatic tissue during tumor progression and metastasis. These results will contribute for a better understanding of the role of Syndecan proteoglycan family and their binding proteins on reactive stroma associated to the prostate cancer development, progression and metastasis.
