Oxidative stress and renal inflammation in rats submitted to ischemia-reperfusion and antioxidant treatment

Abstract

The ischemia and reperfusion (IR) is a condition that occurs during the procedures involved in the transplantation of organs, which consists of one of the causes of acute renal failure of the graft. Despite advances in the area, mortality due to acute renal failure is high. The ischemic process itself is quite harmful, but the reperfusion can lead to a number of complications that can increase tissue and lesions, associated to systemic complications, endanger the patient's life. Objective: To analyze in vivo the anti-inflammatory and antioxidant actions of Brazil nut in rats with renal injury caused by the process of ischemia and reperfusion. Methods: Male Wistar rats, weighing 200 g, will be distributed in four groups (06 rats/group), euthanized after 48 hours of the procedure, as follows: SHAM (surgery without clamping renal artery); SHAM + CP (150 mg of Brazil-nut for 7 days before surgery); IR (IR surgery with clamping renal artery); IR + CP (150 mg of Brazil-nut for 7 days before IR surgery). The following parameters will be evaluated: renal injury and inflammation by histopathological analysis of acute tubular necrosis and neutrophils/macrophages influx and COX-2 expression; oxidative stress by renal expression of iNOS and nitrotirosin. With these considerations, we propose that this study not only will allow to understand the mechanisms involved in the genesis of the lesion by IR, but also alternative targets of potential therapeutic agents in the control of the lesion.
