The recent demonstration of the production of signaling factors reservoirs formed by the self-assembly LbL (layer-by-layer) of polyelectrolyte multilayers has opened new possibilities in tissue engineering. This project proposal basically is aimed at producing the biodegradable and bioadsorble polymeric membranes capable of sustaining the growth of many types of cells. Electrospun membranes will be coated by self-assembly LbL technique through the deposition of pairs of biodegradable polyelectrolytes such as heparan sulfate, pectin, and sodium alginate (anionic), poly-L-lysine and chitosan (cationic). The same LbL technique will also be used to incorporate signaling proteins such as fibroblast growth factor (FGF, acidic and basic), bone morphogenic protein (BMP), vascular endothelial growth factor (VEGF) and platelet-derived growth factor (PDGF) along with the above polyelectrolyte additives in order to enhance bioactivity of the system. These membranes undergo degradation inside the body thereby resulting in slow release of growth factors at membrane/fluid interface. Hence, these loaded multilayered membranes are expected to function as reservoirs of signaling proteins that, upon release, essentially will assist cell events like recruitment, proliferation, differentiation, etc. The combination of growth factors can be used for different types of tissue regeneration. It is intended that such systems act to control the cell proliferation and open up new applications where release of proteinaceous cellular signaling factors will provide specific conditions for differentiation from stem cells.
