Evaluation of the wound healing process in estrogen deficiency rats treated with human OPG-Fc or strontium ranelate: histometric, immunohistochemistry, computed microtomography and confocal microscopy analysis

Abstract

This project aims evaluate the wound healing process after extraction of the incisor of ovariectomized rats that will receive two drugs used in anti-osteoporosis therapy. To develop this project, 168 rats at three months of age will be divided into six groups (SHAM/SAL; OVX/SAL; OVX/RE; SHAM/SH; OVX/SH; OVX/OPG-Fc) which will be initially submitted to ovariectomy or sham surgery. After 30 days, the rats will be submitted to drug treatment by OPG-Fc (10mg/kg/ 2/wk) or strontium ranelate (625mg/kg/day) kept up until the euthanasia. The extraction of the upper right incisor will be held 30 days after treatment initiation and the periods of analysis of wound healing 14 and 42 days after tooth extraction. After euthanasia of animals, according to the proposed experimental periods, the jaw will be removed containing the alveoli healing to be submitted for analysis by computed microtomography; histologic, histometric and immunohistochemical analysis with reaction to OPG, RANKL, TRAP, WNT, OCN and beta catenin; analysis of gene expression by real time PCR and epifluorescence microscopy. The quantitative data will be submitted to normal curve for the definition of a parametric or nonparametric test, with significance level of 5%. (AU)
