Cisplatin's influence on osseointegration of titanium implants: histological, hystometric and immunohistochemical study

Abstract

The aim of this study in animals is to evaluate via comparative analysis by the histological, histometrical and imunohistochemical methods, the peri-implant bone repair against the systemic infusion of Cisplatin. The present study is going to use 60 male rats (Wistar) about 350g weight. Initially the animals are weighed to properly deliver the dose of the anesthetic. The animals are going to be set into two groups, Cisplatin (CIS) and Saline Solution (SS). The titanium implants (2.2mm x 4mm) will be installed on the surgical alveoli, prepared using a 2mm drill coupled on a surgical motor with 45N torque and 1200 rpm speed, under constant irrigation. Group CIS: all the 30 animals will be submitted to the implant installation in an only surgical procedure. Will receive cisplatin via intraperitoneal in two doses, the first one with 5 mg/kg and the second with 2,5 mg/kg, at 15 and 17 days postoperative respectively. At 22, 30 and 60 days after installation the animals will be euthanized. Group SS: all the 30 animals will be submitted to the implant installation in an only surgical procedure. Will receive sodium chloride 0,9% at 15 and 17 days postoperative. At 22, 30 and 60 days after installation the animals will be euthanized. The collected tibiae will be subjected to processing and evaluated via histological, histometric and immunohistochemical analysis, and quantitative data will be analyzed statistically (Bioestat 5.0).
