Atopic dermatitis (AD) is a chronic inflammatory skin condition increasingly common, affecting 15-30% of children in industrialized countries and features a large unmet need for effective therapies. AD is considered a disease with TH2 cell type response characterized by over-expression of proinflammatory cytokines such as IL-4, IL-13 and IL-25. Poumay and colleagues showed that changes in the expression of several genes suggest that keratinocytes RHE respond to depletion of cholesterol as monolayers, however, preserved histology and model barrier function indicates there is some additional activation, probably from the immune system. In this context, this work aims to investigate the tissue response to cholesterol depletion followed by Th2 cytokine exposure in an in vitro model of atopic dermatitis. In the future, this model could be used to evaluate candidate drugs or agents aiming at the prevention or treatment of this pathological condition. (AU)
