INTRODUCTION. Inflammation participates in the different stages of carcinogenesis and tumor progression in the most of the neoplasms. The cancer-associated inflammation, including tumor-associated macrophages, plays different mechanisms depending on the type and frequency of received stimuli, and according of this can display pro or anti-tumor functions. MIF protein acts in the modulation of the tumor immune response and a range of investigations have shown that it can induce the polarization of macrophages located in the tumor stroma to an immunosuppressive and protumoral M2 phenotype. OBJECTIVES. The present study aims assess in vitro the role of MIF protein on the modulation of tumor-associated macrophages and the impact of it on oral squamous cell carcinoma behavior. METHODS. Macrophages from tumor tissue, as well as from healthy subject´s peripheral blood will be obtained and treated in vitro with conditioned media from MIF knockout and wild type SCC15 and SCC25 oral squamous cell carcinoma cell lines. Then the macrophages will be evaluated based on the M1 and M2 phenotype. Functional experiments will be performed to verify the role of the different tumor-associated macrophage´s phenotypes in SCC´s migration, invasion and proliferation.
