Functional detailing of CD99 role in astrocytomas

Abstract

Astrocytomas are between the most common tumors of the central nervous system. According to the World Health Organization, they are classified giving their degree of malignancy in a scale of I to IV and their histological and genetic tumor changes. The grade IV or glioblastoma (GBM) is the most common and malignant. The standard treatment consists of surgical resection of the tumor followed by radiotherapy and chemotherapy with temozolomide. Even with this aggressive treatment, the survival median of patients is approximately 1 year. In order to increase patient survival we aim to combine the standard therapy with other biologically based therapies. Several studies have been developing new strategies in order to identify membrane proteins differentially expressed in several tumor types, including GBM, which could be use as therapeutic targets. Among those targets, we identified CD99, a membrane protein present in hematopoietic cells and other cell types that plays a role in differentiation, adhesion and migration of T and B lymphocytes. CD99 is present in various types of tumors, including sarcoma and GBM. Our laboratory demonstrated an up-regulation of mRNA and CD99 protein in tissue samples and cell line of GBM. In this work, a detailed study of the CD99 functions will be performed by analyzing the transcriptome of GBM cell lines (A172 and U87MG) CD99-silenced and non-silenced, in order to identify new signaling pathways modulated by this protein. The transcriptome analysis will direct the functional studies to be conducted. Additionally, the tumorigenic capacity of CD99-silenced cell lines will be evaluated in vivo.
