Abstract

Periodontitis is a multifactorial chronic inflammatory disease with high prevalent in the population which affects the dental supporting tissues leading to difficult treatment sequels, even with the latest regenerative techniques. Cell therapy seems to be a promising alternative and undifferentiated cells of periodontal ligament and dental pulp have shown great therapeutic potential. Nevertheless, proper induction using biomodifier can increase the potential of cell differentiation. The enamel matrix derivative (EMD) is a type of biomodifier that promotes periodontal regeneration through new cementum formation, but the association of EMD with fresh periodontal ligament and dental pulp cells has not been explored. It has be shown that EMD can enhance odontoblastic and osteoblastic differentiation of dental pulp cells in culture. Thus, the aim of this study is to identify surface markers for mesenchymal cells immediately after obtaining these tissues by flow cytometry, to evaluate bone regenerative potential of ligament periodontal and dental pulp fresh cells and to identify the viability of using biomodifier associations by gene and protein expression analysis. In addition, osteogenic potential of these tissues will be evaluated through an experimental model in rat calvaria associated with EMD. (AU)
