Osteosarcoma is the most common pediatric bone tumor. It is found mainly in the metaphysis of long bones. The current treatment consists on surgical tumor removal and multimodal neoadjuvant and adjuvant chemotherapy. About a third of patients are affected by post-surgical recurrence and distant metastases on lungs and bones. Only 20% of the patients with metastases remain alive after 5 years. Therefore, it is essential to elucidate the molecular pathways involved in the invasive and metastatic process of tumor type. Several miRNAs have been suggested as regulators of the metastatic pathways, including the miR10b. This microRNA is overexpressed in numerous metastatic tumor types. It acts on the invasive processes through an incomplete pairing with the messenger RNA of the HOXD10 gene inhibiting its translation. The HOXD10 is characterized as a general inhibitor of invasion acting on different molecules including the Rho GTPases RhoA and RhoC. These enzymes phosphorylate and activate ROCK1 and ROCK2 kinases that are responsible for the control of the cytoskeleton, an essential mechanism for the cell invasion processes. Furthermore, studies suggest that ROCKS act on cell proliferation, apoptosis and cell cycle control. Many studies have reported overexpression of these kinases in several malignancies, including osteosarcoma. Thus, this study aims to analyze the expression profile of miR10b in osteosarcoma, as well as elucidating its role in the proliferative and invasive processes of this type of cancer. (AU)
