Breast cancer is the second most frequent type of neoplasm worldwide, with higher incidence among women. In this tumor, quantitative and qualitative changes occur in extracelular matrix resulting in remodeling, originated from the activity of proteases secreted by mammary cells in microenvironment. Metalloproteases located in tumor stroma contribute to the tumor invasion, remodeling the extracellular matrix and increasing the malignant potential of epithelial cells. ADAMTS (a desintegrin and metalloproteases with thrombospondin motifs) are known proteases secreted by cells with proteolytic activity on proteoglycans from extracellular matrix. However, data obtained by our group showed that ADAMTS-1 is located predominantly in the cells nuclei and nuclear fraction has proteolytic activity on agrecam. Thus, we are now interested in expanding these results and finding out which is ADAMTS-1 function in the nucleus, whom are its partners and if this protease located within nucleus could exercise regulatory effect on tumor cells behavior.
