Abstract

Microbiota assimilation, high interaction with cations and colloids in soil and low rates of diffusion decrease phosphorus (P) availability to plants. Nitrogen-forms (N) can affect differently P uptake, increasing the release of protons, H+/H2PO4 symporters operation and plasma membrane H+ ATPase and improving P use efficiency. The aims of this study are to investigate if and how N-forms change the expression of phosphate transporters, root architecture, proton-Pi co-transporters operation and the P use efficiency under deficiency and availability of phosphate. Corn plants will be grown in pouches on filter paper and supplied with nutrient solutions varying in their N-source (ammonium or nitrate), P concentration (0, 50, 100, 250 and 1000 ¼M), and pH (4 and 6), in a completely randomized design (2x5x2) and four replications. Root system size and architecture of individual plants will be monitored over three week. At the end of the growth period, plants will be cut and their biomass and P accumulation will be analyzed. Plant mineral composition will be determined by ICP-MS. The activity PM H+-ATPase will be determined colorimetrically. RNA will be extracted from roots and the expression of phosphate transporters will be quantified using rtPCR techniques.
