Abstract

Vitamins are necessary organic molecules for the appropriate functioning of the body. Among the many existing vitamins, the biotin (or vitamin B7) calls attention because, in addition to its functions as an enzyme cofactor in several metabolic pathways, it has high affinity for avidin/streptavidin; one of the largest found in nature. Biotin can be coupled to different types of molecules and can still be recognized by avidin, due to the high affinity constant between them. Thus, this interaction provides great increase in analysis sensitivity; which leads to the development of biosensors and analytical methods for determination of biotin and biotinylated compounds. The use of biological materials such as antibodies, proteins, or enzymes in biosensors becomes often costly and unstable in adverse conditions. An interesting and viable idea is the modification of materials that can mimic the biological interactions of recognition; what has been done successfully by the Molecularly Imprinted Polymers (MIP). The high stability of the polymer allows the modification of them on different materials; as magnetic nanoparticles, increasing the recognition performance of the MIP and, moreover, facilitating the separation of material and solution by applying an external magnetic field. Furthermore, it is also possible to functionalize the surface of the polymer with functional groups, allowing that MIP performs the exclusion of high molecular weight molecules such as proteins; becoming a restricted access material (Restricted Access Media - RAM). Thus, this project aims to develop a selective analysis system through molecularly imprinted polymers immobilized, by the core-shell process, on magnetic nanoparticles and modified with hydrophilic groups to create a restricted access, forming a magnetic RAMIP (Restricted Access Molecularly Imprinted Polymer) for biotin analysis. For this, the polymer is synthesized by different routes (semicovalent or non-covalent) and to provide the hydrophilic surface will be modified with glycerol dimethacrylate (GDMA) and/or hydroxy methyl methacrylate (HEMA). After properly characterized and with maximized parameters (such as retention efficiency and selectivity), the polymer with the best results will be applied in a lateral flow device, which will be optimized and evaluated on samples of blood and urine containing biotin, aiming subsequent application in the determination of biotinylated biological molecules in this type of samples.
