Abstract

The reduction of the viable bacterial load after endodontic disinfection procedures is a key element for treatment success. Since ribosomal RNA(rRNA) indicates microbial viability, molecular approaches based on rRNA are choice methods to assess the antimicrobial effectiveness of endodontic procedures. The objective of the present study is to assess the viability of as-yet-uncultivated bacteria after chemo-mechanical procedures during endodontic retreatment using rRNA-based molecular methods. Samples from18 teeth with persistent/ secondary intra-radicular infection before (S1) and after(S2) chemo-mechanical preparation will be used in this study. cDNA, which was synthesized from RNA, will be subjected to Real-Time Polymerase chain reaction (qPCR) with primers complementary for Bacteroidetes oral clone X083e Olsenella uli 16S rRNA sequences. The bacterial reduction will be analyzed by statistical tests.
