Fluoride influence on the membrane protein expression in mesenchymal cell

Abstract

The recent development of proteomics techniques has allowed the complete protein profile analysis of biological systems allowing a better understanding of the body's normal physiology and mechanisms of disease, the discovery of biomarkers for early disease detection, identification and discovery of new therapies drugs. This study uses proteomics techniques to analyses the effects of fluoride (F) on the cell viability and expression of membrane proteins of mesenchymal cells from two strains of mice with different sensitivities to dental fluorosis. To this end, we analyzed the effect of different concentrations of NaF on the viability of MSCs obtained from the animals A/J (susceptible to fluorosis) and 129P3 /J (resistant fluorosis) by MTT assays and quantification of total protein; and also assess the effect of different concentrations of NaF in the expression of membrane proteins from the cell membrane in mesenchymal cell lineages A / J and 129P3 / J. These mesenchymal cells (9th passage) will be plated on the ±-MEM / 10% FBS and will be treated with NaF concentrations of zero, 10-5, 10-3 M, at 6pm exposure time. The membrane proteins of the mesenchymal cells are isolated by ultracentrifugation. After extraction of membrane proteins, the free proteomic markers is performed using n-LS-ESI-MS / MS (AU)
