Abstract

This project is part of Validação química e farmacológica de extratos e princípios ativos de espécies de Copaifera FAPESP 2011/13630-7 and a continuation of the project Avaliação do processo de cicatrização de lesões cutâneas utilizando extrato hidroalcóolico e óleo de copaíba FAPESP 2012/15092-5. The tegumental system is highly complex, essential to maintaining the integrity of the body, being a protective differentiated barrier. The discontinuity of this system promotes the healing mechanism, physiological process that, after injury, for total epithelial renewal, can be didactically divided into interdependent and overlapping phases: the inflammatory phase, whose main events are hemostasis and inflammation, proliferative phase, in which there is the formation of granulation tissue and phase of remodeling of extracellular matrix. Due to the possibility of some kind of physical disability, psychosocial consequences or even lead to death, the skin lesions have great importance in the investment of public resources. The Copaifera langsdorffii Desf. (Leguminosae) is a native tree of tropical regions of Latin America and West Africa witch, according to popular knowledge, has anti-inflammatory, antimicrobial and wound healing activities. Therefore, this project aims to investigate the effect and mechanisms involved in the healing action of ointments of hydroalcoholic extract of leaves and oil resin of bark of Copaifera langsdorffii in skin wounds of rats. Male Wistar rats will be randomly divided into four experimental groups (n = 8): base lanete ointment (negative control), Collagenase ointment (positive control), hydroalcoholic extract 10% ointment and oil resin 10% ointment. The animals will be anesthetized and then dorsal lesion will be made with a punch of 2 cm of diameter. The lesions will be treated and measured once a day using a digital caliper for 3, 7 and 14 days (according to the phases described in the literature). After euthanasia, the region of the wound will be designed to histological analysis (count of inflammatory cells, inflammatory score, morphology and deposition of collagen fibers), morphometric (height of the epithelium regeneration, thickness of epidermis and dermis) and immunohistochemical (metalloproteinase-2, metalloproteinase-9, EGF and NF-kappa²), determination of collagen, and molecular analyzes (qPCR and RT-PCR) for collagen I and III, and growth factors such as TGF-²1 and EGF. Antioxidant activity will be measured (reduced glutathione, glutathione reductase and peroxidase, superoxide dismutase and myeloperoxidase), anti-inflammatory activity by measuring cytokines in blood serum by ELISA method (TNF-±, IL-1², IL-6 and IL will also be determined -10).
