Effectiveness evaluation of a gel with low concentration fluoride supplemented with sodium trimetaphosphate on demineralization enamel dental: study in situ

The proposal of this study is to evaluate the ability of a topical gel neutral pH highly concentrated fluoride supplemented with sodium trimetaphosphate (TMP) in situ reducing demineralization of the dental enamel. Will be performed 5 experimental periods with duration of seven days each, and washout of 7 days amongst them, one blind, crossover study. Bovine enamel blocks (4 mm x 4 mm; n = 240) are prepared from bovine incisors and selected from the initial surface hardness (SH). Volunteers (n = 12) use intra-oral palatal appliances containing 4 enamel fragments, which will be submitted to the following treatments: 1) Toothpaste Fluoride 1100 µg F/g (DF) (1100); 2) Gel without F/ TMP (Placebo); 3) Gel 4500 µg F/g + 5% TMP (4500 5%TMP); 4) Gel 9000 µg F/g (9000) and 5) Gel 12300 µg F/g (Acid Gel). After topical application of the gel or fluoride toothpaste for 1 minute, 2 blocks are removed for analysis of calcium fluoride concentration formed in the enamel (CaF2) (n = 120). The cariogenic challenge will be produced by 30% sucrose solution, 6x/day. After the experimental period, will be determined the final surface hardness (SH1), percentage of surface hardness change (%SH), hardness in longitudinal section (”KHN) and calcium fluoride content (CaF2), retained on enamel (n = 120). Statistical analysis will be considered as variables the values of %SH, ”KHN and CaF2, formed and retained on enamel and as a variation factor, toothpaste and experimental and commercial gels. A more appropriate statistical test (p <0.05) will be used after analysis of normality and homogeneity of data. (AU)
