Abstract

Topical application of nitric oxide (NO)-releasing hydrogels may prevent or even reverse the consequences of endothelial dysfunction, such as diabetic leg ulcers and double network hydrogels (DNH) may increase the potential applications of the conventional NO releasing hydrogels due to their enhanced mechanical properties. The main objectives of this project are the preparation of new DNH hydrogels as platforms for the thermal and photochemical NO release for biomedical applications. Two main platforms will be developed: DNH composed of poly(vinyl alcohol) (PVA) interpenetrated with hydroxypropylmethylcellulose (HPMC), both crosslinked via esterification with mercaptosuccinic acid and DNH composed of PVA functionalized with thiolactic acid (TLA), interpenetrated with a network of Pluronic F127 micelles. Both platforms will be subjected to further S-nitrosation of the -SH moieties yielding -SNO-functionalized DNH. The DNH will be characterized regarding their viscoelastic behavior by oscillatory rheology, thermal properties by differential scanning calorimetry and diffusion properties using Franz diffusion cells. The changes in the microstructure of the DNHs under stretching and compression conditions will be investigated by Small Angle X-ray Scattering (SAXS) at the Brazilian Synchrotron Light Laboratory (LNLS), Campinas, SP. The kinetics of thermal and photochemical NO release from the DNH will be characterized by chemiluminescence. The potential applications of the new NO-releasing DNHs for topical use and for use as artificial cartilages will be evaluated. (AU)
