Abstract

The mTOR signaling pathway has been associated with several diseases and metabolic disorders in humans, including obesity, diabetes and several types of cancers. The S6Ks proteins have shown an important role in mTOR signaling, acting as effectors of this pathway. However, the functional differences between S6Ks isoforms is still an open question. The family of S6Ks is composed of two genes, S6K1 and S6K2, and the following isoforms will be used in this study: p70-S6K1, p85-S6K1 and p54-S6K2. The aim of this study is to functionally characterize different interacting proteins of S6Ks isoforms. Therefore, p70-S6K1, p85-S6K1 and p54-S6K2 will be overexpressed in human HEK293 cells, immunoprecipitated and different interacting partners of these isoforms will be identified by mass spectrometry. In addition, we intend in this project to identify the role of the interactions between these proteins and S6Ks, looking for phosphorylation sites of target proteins, S6Ks activation levels and the biochemical nature of the interaction that occurs between them.
