Abstract

Prostatic cancer is the main type of tumor in male worldwide, and the frequency of relapsing and metastatic diseases demands the development of new approaches for diagnosis and therapy. Considering this cenario, this project aims to develop nanocarriers to transport antineoplasic agents into tumor cells. For this purpose, carbon nanotubes will be functionalized with polyethylenimina (PEI) for incorporation of paclitaxel, a first line medication for prostate cancer. Then, nanotubos will be recovered with anti-prostate specific membrane antigens (anti-PSMA) to address then for PSMA+ tumor cells. In addition, particles will be conjugated to fluorescein isotiocianate in order to follow the cell adhesion/internalization. Nanocarries will be tested in vitro for: a) specificity for PSMA+ tumor cells; b) uptake by target cells, and c) cytotoxicity of paclitaxel immobilized on nanotubos. Target specificity will be tested by exposition of nanocarrier to LNCap cells (PSMA+ cellprostate cancer), HCT-116 cells (PSMA-colon cancer cells), peripheral blood lymphocytes and monocytes. Uptake of carriers by these targets will be analyzed by imunofluorescence microscopy and flow cytometry. Cytotoxicyty of immobilized paclitaxel will be checked by in vitro exposition of target cells to nanocarriers and further analysis of ability to reduce methyl-tetrazolium salt (MTT). (AU)
