Identification of mesenchymal stem cells from human periodontal ligament that have high osteoblastic/cementoblástico potential

Abstract

Studies with the aim of evaluating the potential of mesenchymal progenitor cells in the periodontal ligament regeneration of periodontal supporting tissues has shown that these populations are highly heterogeneous, with no a cellular phenotype defined as the most favorable for this regenerative process. Considering the new formation of cementum a crucial process for the restoration of the connective tissue supporting the teeth affected by periodontal disease, the present study aimed to isolate and characterize homogeneous populations of undifferentiated mesenchymal cells isolated from human periodontal ligament with high potential for differentiation osteoblastic/cementiblástica. Using the technique of cloning cylinder, cell clones (homogeneous populations) are isolated from mesenquimias three populations of stem cells of the periodontal ligament and characterized as: a) potential for osteoblast/cementoblástica differentiation (Alizarin test "red" and expression of genes for RUNX2, and ALP by qPCR OCN); b) ability to differentiate into cells similar to adipocytes (Oil red O staining and expression of genes for LPL and PPARg by qPCR); c) proliferative capacity curve (cell growth); d) expression of STRO-1 protein by immunofluorescence.
