Zinc modulation in the diet during the prostatic morphogenesis and the consequences on the development and glandular differentiation

Abstract

Zinc is an essential trace element for living organisms due to its numerous biological activities. It acts as a growth factor and immunoregulatory and presents antioxidant and anti-inflammatory properties. To be involved in many aspects of cellular physiology, including structural and functional conditions, the change in their levels during normal sexual development can modulate a series of events that begin in fetal life and continue after birth. Thus, modulation of environmental factors such as diet, may lead to significant changes in the development, including on the prostate gland morphogenesis. This project aims to investigate whether exposure to diets with disabilities or zinc supplementation during pregnancy, lactation and juvenile interfere in prostate development in pubertal and adult stages. This project is divided into two steps: Step 1: Step fulfilled by the student during his Scientific Initiation with Exchange PIBIC; Step 2: Step to be fulfilled in the proposed period. Step 1: Pregnant rats of the Sprague-Dawley strain (n = 10 / group) were exposed to standard diet (AIN 93G) with normal levels (0,012mg / g), disability (0.005 mg / g) or supplementation (0,18mg / g) zinc. The pregnant mice were given the experimental diets ad libitum from gestational day 10 to postnatal day 21 (PND 21). After weaning, male offspring were divided into three groups (20 animals / group) receiving the same diets until PND 53 (G1: standard diet; G2: zinc-deficient diet; G3: diet with zinc supplementation). In DPN53 and DPN115 the animals were weighed and euthanized by decapitation, proceeding to collect blood for hormone dosage. The ventral prostate was weighed, the right hemilobo for the histological routine and the frozen left hemilobo. Histological sections of the right hemilobo were stained with H & E, Picrosirius and Reticulin Gomori and intended for stereological analysis. Immunohistochemical reactions for the androgen receptor (AR) and Ki67 were performed. Step 2 Proposed Project: The cell proliferation (Ki67), cell death (apoptotic phenotypes) and the proportion of stromal collagen will be quantified by means of histological sections. The hemilobos prostatic collected and frozen left will be allocated to protein extraction and quantification by the Bradford method. To analyze the activity of metalloproteinases 2 and 9 will be held the test zymography for digestion of gelatinases. The AR protein (androgen receptor) and SIRT1 (cellular stress) will be examined by semi-quantitative Western Blot.
