Assessment of renal protection from fenoldopam in experimental model of renal ischemia and reperfusion in rats under inhalation anesthesia with isoflurane

Abstract

Background and Objective: Renal ischemia/reperfusion injury is a major cause of acute renal failure is a common phenomenon in the perioperative period and in critically ill patients hospitalized in intensive care units. The restoration of blood flow may aggravate the injury by the mechanism of ischemia and reperfusion (I/R). The aim of this study is to evaluate the infusion of fenoldopam in rats submitted to the I/R process in an experimental model. Method: 30 Wistar rats will be selected and randomly divided into three groups. Ischemia Group (right nephrectomy, infusion of saline solution and left renal I/R, n=10), Fenoldopam Group (right nephrectomy; fenoldopam infusion, 5¼g.kg-1.min, intravenous infusion one hour before the ischemic procedure); and left renal, n=10) and Sham Group (laparotomy and saline infusion, n=10). The animals will be anesthetized with isoflurane. During the experimental procedure, the following criteria will be evaluated: weight, heart rate (HR), systolic blood pressure (SBP), diastolic blood pressure (DBP) and mean arterial pressure (MAP), intraoperative temperature (T) at M0 moments (initial monitoring), M1 (after stabilization), M2 (after ischemia) and M3 (after reperfusion). Plasma levels: NGAL, IL-18, KIM-1 and creatinine will be held at moments M1, M2, M3, M4, M5, M6 and M7 (24 hours after the beginning of the experiment). The animals will be placed in metabolic cages for urine collection between M3 and M7.The kidneys will be analyzed histologically for determination of acute tubular necrosis and evaluation of necrosis and apoptosis by flow cytometry.
