In leishmaniasis it is known that for the resistance and susceptibility to infection the cellular immune response is considered most important. However in the initial phase non-specific factors are considered fundamental in determining the course of the disease. Besides the complement system, macrophage reactive oxygen and reactive nitrogen species, neutrophils and eosinophils, other factors have being considered including the Insulin-like Growth Factor-I. Great interest on the study of IGF-I in immunity in leishmaniasis was originated from the works done by Prof. Dr. Hiro Goto where it was shown that extrinsic IGF-I favors the proliferation of Leishmania and progression of the parasite infection, with decreased production of nitric oxide and arginase activation of macrophages and the parasite. In the present project we will study the influence of IGF-I on the parasitism of BALB/c mouse peritoneal macrophages using Leishmania (Leishmania) infantum. Similar studies are in progress with Leishmania (L.) major and Leishmania (L.) amazonensis but knowing that there are considerable biological differences among different Leishmania species, we will develop the studies now with L. (L.) infantum. The results from this study will contribute to the understanding of the differences in the behavior of infections caused by different Leishmania species and of the pathogenesis of visceral leishmaniasis.
