Abstract

Biological markers are useful tools to assist the management of species and ecosystems, with proven efficacy in several studies. Among the highlights the micronucleus test, which is fast, cheap, and easy to perform, effectively already tested in various situations. Mangroves are Permanent Protection Areas (PPAs), being relevant environments to many aquatic and terrestrial animals, as well as to adjacent ecosystems. In this sense, will be established basal levels of micronucleated cells in two species of brachyuran crustaceans, Aratus pisonii (arboreal crab) and Goniopsis cruentata ("maria-mulata" crab) in a Conservation Unit (CU), the Mosaic of CUs Juréia-Itatins. Fifteen males specimens in the intermolt stage will be collected of each species at three sampling points (n=5/point), which occurs in two seasons (summer and winter) to test the possible effect of seasonality on this biomarker. The hemolymph of the animals will be collected for making smears, that will be fixed (Carnoy's solution), colored (Giemsa solution 2%) and submitted to optical microscopy for counting the frequency of micronuclei per 1,000 cells analyzed (MN0) using specimen as replicates. The results for the specimen shall be submitted to statistical analysis (parametric or nonparametric) for comparison of central tendency measures (means or medians) obtained for each species between seasons. The results will allow a better understanding of the biomarker response, checking a possible difference between these species of crabs and the effect of the seasonality. New information will be provided the use of this tool to establish the state of conservation and management of these species in the mangrove protected areas. (AU)
