Paternal exercise modulates offspring glucose homeostasis: The role of DNA methylation.Chronic complications associated with diabetes mellitus result in increasing morbidity, reduce life expectancy, and cause enormous health cost. There is strong evidence that obesity and physical inactivity are the main nongenetic determinants of diabetes. It is also known that alteration in epigenetic mechanisms may be associated with this disease. The effects of maternal nutrition on the metabolic programming of the offspring are well established and, recently, it has been demonstrated an important biological role of the father in this context. Although the beneficial effects of regular exercise are well recognized, little is known regarding exercise performed by the male progenitors in the next generations. Thus, we will analyze the effects of chronic endurance exercise, performed by male progenitors, on: a) glucose homeostasis; b) the molecular and functional mechanisms of insulin secretion in isolated pancreatic islet, c) the insulin sensitivity in skeletal muscle from offspring fed a control or high fat diet and, d) the possible involvement of epigenetic modulation on these processes by either diet or exercise. The last objective will be conducted at the laboratory of Dr. Klaus Kaestner, at the University of Pennsylvania, where we intend to learn the techniques involved in DNA methylation analysis.
