The project to be developed by the student is part of a broader project that aims at the development of immunosensors from monoclonal antibodies immobilized on polymeric matrices in nanostructured films (Project Universal 480896/2013-5). The proposal is based on the immobilization of monoclonal antibody anti-FDP (fibrin degradation product) on silk fibroin in LbL films for the diagnosis of venous thromboembolism. Initially will be necessary to standardize the method of immobilization of the biomolecules and investigate the molecular mechanisms involved in the interaction between the silk fibroin and anti-PDF. These studies will be conducted using spectroscopic techniques, UV-visible and fluorescence. The response of the immunosensor will be investigated by antigen-antibody interaction, i. e. the D-dimer, which is the fibrin degradation productf, and anti-FDP, which will be monitored with measurements of electrical impedance and/or electrochemical.
