Abstract

This study will aim to evaluate the level of Babesia bovis and Babesia bigemina,infestation by Rhipicephalus (Boophilus) microplus and gene expression of interferon gamma (IFN-g in beef cattle Canchim (5/8 Charolais + 3/8 Zebu)steers grazing naturally infested by ticks. shall be used 50 cattle belonging to the flock Embrapa cattle Southeast, located in the city of São Carlos-SP, a region previously characterized as stable endemic for bovine babesiosis. Shallbe held 8 hemoparasites measurements of blood samples by means of the technique of real-time PCR (qPCR), one in each season (spring, summer,autumn and winter), in a period of 24 months. Consideration will also, the gene expression gamma interferon (IFN-g) in animal blood, once in each season. However, the study of IFN-g is after the analysis of the level of babesias infection by qPCR, aiming thus evaluate only the animals from extreme resistance / susceptibility. The degree of tick infestation is evaluated by scores of females with size greater than or equal to 4.5 mm in diameter present on the left side of each animal, this procedure will be done simultaneously with blood samples for testing of qPCR. For the analysis of gene expression data, the methodology used is that described by Livak and Schmittgen et al. (2001). Results of counts of ticks, the number of copies for Babesia spp., and expression of IFN-g be evaluated statistically using the MIXED procedure of the SAS statistical package (2001), in order to verify the occurrence of associationsto characterize animals as resistant and sensitive.
