Abstract

During the inflammatory response, biochemical mediators are released locally in order to stimulate the humoral and cellular immune response. Through the action of the enzyme cyclooxygenase (COX) and lipoxygenase (LO), structural changes occur in the chain of arachidonic acid, leading to the synthesis of these mediators. The objective of this study is to investigate the role of inhibitors of the enzyme 5-lipoxygenase in the treatment of experimental periapical lesions in C57BL / 6. 54 will be used C57BL / 6 mice, six weeks old, weighing from 20 to 22 grams are inoculated in which the root canals of the first molars a solution containing bacterial lipopolysaccharide from E. coli (1.0 mg/ml). After 30 days of induction and as well as the corresponding experimental periods of 7, 14, 21, and 28 days, the animals will be euthanized and the blocks containing the teeth and bone are removed for extraction of total RNA. Then there will be the evaluation of gene expression by a reverse transcription-polymerase chain reaction in real-time (qRT-PCR). The values of the relative expression of each mRNA for each group are compared by analysis of variance (ANOVA) followed by two-way Bonferroni post-test or one-way ANOVA followed by Dunnett's post-test (= ± 0.05).
