Abstract

The intracellular calcium (Ca2 +) is essential for the maintenance of homeostasis and cell functions in the nervous system (NS). Deregulation of Ca2+ concentration has been increasingly related to injuries, and degenerative diseases. The receptor of inositol -1, 4,5 triphosphate (IP3R) is involved in this regulation but their role has not yet been clarified. The aim of this study is to evaluate the distribution of IP3Rs in the retina control and after different times of injury, and describe the specific cell type expression. Firstly, we will determine the modulation of gene expression and protein levels of IP3R1, IP3R2 and IP3R3 after mechanical trauma in the retina, this model was well established in our laboratory, which allows the visualization of the focus, shadows and adjacent areas to the lesion. With the combination of techniques such as polymerase chain reaction in real time (Real-Time PCR) and immune-histochemistry, we want to evaluate the spatiotemporal expression of IP3Rs after injury. To determine the role of these receptors in the secondary neuronal loss we will use pharmacological inhibitors combined with methods for assessing cell death, such as terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL), lactate dehydrogenase (LDH). In resume, we hope to clarify aspects of the plasticity of IP3Rs during neurodegenerative processes.
