Microtomography and histomorphometry evaluation of bone defect repair treated with porous HA/TCP and anorganic bovine bone in rabbit skull defect

This study aims to compare the bone formation in critical size defects in rabbit skull treated with a) porous HA/TCP and b) anorganic bovine bone (Bio-Oss). In 12 white rabbits (Oryctolagus cuniculus), bilateral parietal defects of 9-mm diameter will be created and the left defect will be filled with 200 mg of HA/TCPp and right defect with 200mg of Bio-Oss. After 4 and 8 weeks (6 animals per period), the skulls will be collected and fixed in 10% formalin. The pieces will be analyzed by microtomography to assess the volume of the bone and graft comparatively between groups and experimental periods. Subsequently, the pieces will be demineralized and subjected to histological standardized procedures. In semi-serial cuts of 5-mm sections will be stained by HE and verified the histological tissue response to biomaterials and the analysis descriptive will be performed to verify tissue response to implanted materials and morphometric to obtain the percentages of new bone, connective tissue, biomaterial and myeloid tissue. All data will be compared between different periods and groups by analysis of variance (ANOVA) or Kruskal-Wallis test, depending on the normality of the distribution of data and and Tukey's HSD Post-hoc test. Will be adopted the significance level of 5%.
