Functional characterization of non-coding RNAs associated with IS1341 transposons in Halobacterium salinarum

Abstract

Mobile Genetic Elements (MGEs) are important players in the plasticity and evolution of genomes. Their impacts are diverse, ranging from antibiotic resistance in microrganisms to human disease. One important class of MGEs are the Insertion Sequences (ISs). These elements are found mainly in bacteria and archaea and present a huge diversity of families and mechanisms. An interesting family is the IS200/605, this family is divided in 3 subgroups: IS200, IS605 and IS1341. Another interesting aspect is the utilization of single-stranded DNA as a substract for transposition. In a recent study by Gomes-Filho et al., 2015, it was shown the presence of sense overlapping transcripts (sotRNAs) at the 3' end of IS1341 genes in archaea and bacteria. These RNAs present a common motif called UUCA tetraloop, which was also found in an intergenic non-coding RNA. Overexpression of RNAs containing this motif in H. salinarum improved it's growth. Thus, to continue the functional characterization of these newly identified molecules we will apply phenotypical assays and molecular techniques in order to fully understand the function and mechanisms of these RNAs.
