Abstract

Tumor necrosis factor (TNF), is a proinflammatory cytokine and some studies demonstrate that TNF-alpha acts as a potent inhibitor of gastric secretion after Helicobacter pylori infection, contributing to the development of gastric diseases, and acting as the most powerful mediator of acute inflammatory response against Gram-negative bacterium. In addition, Helicobacter pylori (H. pylori) a Gram-negative bacterium, which persistently colonizes the gastric mucosa of humans and plays an important role in gastrointestinal diseases and appears to induce the production of TNF-alpha, contributing to the development of peptic diseases. In view of the involvement of TNF-alpha and the presence of H. pylori in the development of gastric diseases, this project aims to characterize the promoter region polymorphism -308 (rs1800629) of TNF by PCR-RFLP and analysis the gene expression of the TNF-alpha gene in 125 samples from gastric biopsies of adults patients with peptic symptoms and/or gastric cancer, in order to correlate genotypes and the genes expression of TNF-alpha with the presence of H. pylori and some virulence markers previous detected. The development of this approach may open new perspectives for the characterization of factors involved in peptic diseases, gene regulation, as well as to elucidate the interaction among genes involved in gastric diseases, improving the understanding of the pathophysiological mechanisms of these diseases.
