The present project proposed for the Doctor degree of Luciana da Silva Amaral, aims to develop and apply analytical methodologies to verify the influence of nitrogenated additives on the secondary metabolism of the endophytic fungi coded as NICL3 and NICL5, both belonging to Xylaria genus. Previous studies showed that NICL3 produces eremophilane sesquiterpenes and NICL5 produces mainly cytochalasans. Many eremophilane sesquiterpenes are known for their anti-HIV and bactericidal activities. Cytochalasans have attracted great deal of attention, due to their unusual molecular structures, which are characterized by a highly substituted perhydroisoindole moiety linked to a C16 - C18 polyketide ring system. Depending on the amino acid incorporated into the polyketide chain, the hydrogenated isoindolone either bears a benzyl group (cytochalasins), a p-methoxybenzyl group (pyrichalasins), an indol-3-yl group (chaetoglobosins), a 2-methylpropyl group (aspochalasins) or a methyl group (alachalasins). Due to the high structural diversity, the cytochalasans has drawn much attention due to their pronounced biological activities, mainly the ability to cause cells to extrude their nuclei, inhibition of HIV-1 protease, antibiotic, among others.In the present project the effect of different nitrogen source, such as amino acids and nitrogenated inorganic nutrients, on the secondary metabolism of the endophytic fungi, will be studied using liquid chromatography coupled with mass spectrometry techniques.
