Abstract

The kallikreins are serine proteases involved in many physiological functions, levels changes of kallikreins 6 and 8 in the tissues or serum implies in several diseases as some types of cancers, neurodegenerative disease and neuroinflammatory disorder (SCARISBRICK et al., 2002; BLABER et al., 2004). Thus, the aim this work is to evaluated the kallikrein 6 and 8 expression and levels and to evaluated expression of tissue kallikrein inhibitor (kallistatin) as well as the levels of plasmatic kallikrein inhibitor (C1INH) through Wester Blot, immunohistochemistry, Real Time PCR and ELISA assays in the hippocampus and serum of patients with temporal lobe epilepsy submitted to corticoamigadolohippocampectomy for seizure control. Selected patients undergo a detailed history, with video-EEG monitoring and study of magnetic resonance imaging. The control tissues will be obtained from autopsies performed in the service of verification of death of the Hospital das Clinicas (less than 5 hours post mortem). In this work we will also use hippocampus from rats submitted to the epilepsy model of pilocarpine to analyze the kallikrein 6 and 8 expression, and the expression of tissue kallkirein inhibitor, the kallistatin, by Wester Blot, Real Time PCR and immunohistochemistry assays. Animals will be divides according to the periods of the epilepsy model in: acute, silent and chronic phases and their respective controls received only saline. (AU)
