Abstract

The intestinal tract contains one of the most complex ecosystems of any environment on the planet, characterized by high diversity and richness. The intestinal microbiota plays a critical role in health and disease. Studies using next generation sequencing to characterize the equine intestinal microbiota are rare and the investigation of the effects of training and ergogenic supplementation has not been performed to date. The objective of this study is to use next generation sequencing to investigate changes on the fecal microbiota of horses before and after training, receiving or not supplementation with oral chrome or carnitine. In order to do that, twenty two "Mangalarga Machador" mares receiving the same diet will be randomly assigned into four groups: control not trained nor supplemented (GC, n=6), trained but not supplemented (GD), trained and supplemented with 10 g of oral carnitine daily (GCAR, n=6) and trained and supplemented with 10 mg of oral chrome daily (GCR, n=5). Horses will be trained on a treadmill three times per week and on an automatic horse walker twice per week for six weeks using medium exercise intensity (70% of the lactate threshold). Fecal samples will be collected before and after the training season. Approximately 10g of feces will be collected by rectum and immediately frozen at -80ÚC until DNA extraction. Sequencing of the V3-V4 region of the 16S rRNA gene will be performed after PCR amplification with the 2x250 MiSeq Illumina sequencer. The software QIIME will be used for data cleaning and statistical analysis.
