Seminal evaluation, determination of concentration and profile of proteins present in seminal plasma of two species of brown brocket deer (Mazama gouazoubira and M. nemorivaga) maintained in captivity

The determination of profile and concentration of the seminal plasma (SP) protein will contribute to understand the molecular mechanism of regulation of physiological reproductive parameters and the interactions between semen and sperm surface and, assist in the comprehension of natural reproduction of each species, their reproductive cycle and differences of seminal composition between species. The results may contribute to conservation techniques development. There are few reports on deer's SP protein composition what difficult their conservation, thus, in this study the seminal characteristics of two species of Brown Brocket Deer, Mazama gouazoubira and M. nemorivaga, will be evaluated. In this way, two samples of each animal will be collected with an interval of 20 days. Immediately after the collection volume, color, concentration, motility and vigor of the samples will be evaluated and posteriorly the SP will be centrifuged and stored. The total plasma protein concentration will be determined using the procedure described by Hartree (1972). The protein profile determination will be conducted through a polyacrylamide gel electrophoresis in presence and absence of denaturing agent, sodium dodecyl sulfate. The results will be subjected to normality and homoscedasticity verification and posteriorly to the variance analysis with probability level of 5% and, the significant differences between the concentrations will compared by the Tukey test.
