Abstract

The angiogenesis consists in the formation of new blood vessels from the pre-existenting vasculature and the vascular endothelial growth factor (VEGF) is a key regulator in this process, being a survival factor for endothelial cells, stimulates the origin of the new microvascular network. The process of angiogenesis is extremely necessary for successful transplantation, regeneration and repair of damaged tissue recovery after ischemia and wound healing. Thus, understanding of enhancing growth of new microvascular system is essential for new therapeutic strategies. Recent studies have shown that the protein annexin A1 (ANXA1) is involved in the regulation of inflammation, apoptosis, cell growth, differentiation, and also the balance between physiological and pathological angiogenesis, and can act as a pro-angiogenic factor. With these considerations, our aim is to investigate the role of this protein in the process of angiogenesis, using experimental models in vivo and in vitro induced by VEGF. The effects of ANXA1 on the kinetics of formation of new vessels in vivo microcirculatory network, will be investigated in an experimental model of dorsal chambers implatation in Balb/c wild and ANXA1 knockout mouse. In investigations evaluate in vitro, in cell line of human umbilical vein endothelial cells (HUVEC), the effect of protein ANXA1 in the following aspects: cell viability, proliferation, migration, tube formation, adhesion molecule expression, adhesion and quantification of ANXA1 endogenous. Although some investigations have identified the involvement of ANXA1 in the regulation of angiogenesis, the exact role in the physiology is not fully understood. Thus, our studies, taken together, might reveal important functions of ANXA1 in this process that involves proliferation and differentiation of endothelial cells and extracellular matrix surrounding cells.
