Abstract

This study aims to clarify mechanisms of copper toxicity, accumulation and tissue distribution in the fiddler crab Uca rapax (Crustacea, Ocypodidea), a semi-terrestrial crustacean that inhabits a wide range of salinities. Our ultimate goal is to develop a Biotic Ligand Model approach and compare different pathways of metal uptake over a range of salinities after exposure to waterborne and dietary copper. Crabs will be collected on Key Biscayne, Miami, Florida, transported to the Laboratory of Environmental Physiology & Toxicology, at the University of Miami, Florida, where they will be exposed for 10 days to waterborne Cu in contaminated food. Toxicity tests will be performed at different salinities (<0.5, 25 and 60 S) together with measurements of physicochemical parameters and total and dissolved copper concentrations in the experimental media. Thus specific objectives are to examine the influence of salinity on waterborne and dietary copper toxicity, accompanying hemolymph osmolality and ion (Na+ and Cl-) concentrations the gill Na+/K+ ATPase and carbonic anhydrase activity accumulation of copper in different tissues.
