Abstract

Skeletal muscle is highly specialized in the production of force and motion, can adapt to different types of stimuli such as increased demand for mechanical, hormonal status and injury. The injury leads to regenerative response aimed at tissue repair. The regenerative process is extremely important to maintaining the quality of skeletal muscle tissue not only after acute injury but also to deal with micro injuries that occur in daily life activities. Thus the knowledge of the mechanisms cellular / molecular involved in the regenerative process are also important. Studies of another ongoing project in our laboratory show that MuRF1 and MuRF2, has an important role during muscle regeneration (results presented in the introduction), that go beyond their well-known roles as E3 ligases in the process of muscle atrophy . We know that these two MuRFs are strongly induced during regeneration by an unknown mechanism. Accordingly which fits this proposal, we hypothesized that certain comprises microRNAs are capable of inhibiting MuRF1 and MurF2 after injury and that these microRNAs are suppressed, allowing the overexpression of MuRFs. Initially seek in the literature and also in silico microRNAs potentially involved with inhibition of MuRFs. Then will elect two microRNAs potentially inhibiting MuRF1 and MuRF2 in the context of skeletal muscle regeneration. Investigate the expression of these microRNAs in vivo and in vitro, experimentally verifying if MuRFs are effectively targets of these microRNAs. Finally will induce rising levels of these microRNAs in vivo and in vitro in order to understand their impact on the regenerative process. (AU)
