Abstract

Obesity and insulin resistance are related to glucose and fat acids metabolism hepatic alterations. The insulin resistance promotes transcription factors activation that elevates gluconeogenesis and fatty acids de novo hepatic production, inhibit glycogen synthesis and reduces glucose and fatty acids oxidation. This effect enhances free fatty acids circulation levels and induces triacylglycerols ectopic deposition on hepatic parenchyma, impairing obese patient healthy quality. Recent studies demonstrate that palmitoleic acid (or palmitoleate) have anti-inflammatory properties and operates on muscle and liver as adipokine, which improve insulin resistance and enhance liver lipids oxidation. These effects could be associates to PPAR-alpha, a nuclear receptor that inhibits pro-inflammatory pathways and enhances mitochondria and particularly peroxisome beta-oxidation. Thus, our objective are investigate palmitoleate supplementation effect, as well as PPAR-alpha influence on liver inflammatory process, on high fat diet insulin resistance and on hepatic lipids synthesis and oxidation. Therefore, we will investigate in C57BL/6J and PPAR-alpha knockout (PPAR-alpha(-/-) mice the palmitoleate supplementation effects on peripheral and hepatic insulin resistance, we will determine the palmitoleic acid effect on liver inflammatory process, will evaluate lipids hepatic metabolism by the novo synthesis, mitochondrial and peroxisomal beta-oxidation, krebs cycle and electron transport chain, finally, we will confect liver histological slices to determinate inflammatory cells and the accumulation of lipids in the liver,verifying palmitoleate effect on hepatic steatosis. Therefore, this project will be important to understand the mechanisms involved in liver palmitoleic acid action, elucidating obesity, diabetes mellitus type 2 and associates conditions related questions.
