Abstract

Tauroursodeoxycholic acid (TUDCA) is formed from the conjugation of bile acid ursodeoxycholic (UDCA) with the amino acid taurine (Tau). Besides the well-described actions on gastrointestinal function, the TUDCA contributes to the restoration of glucose homeostasis in Type 2 Diabetes Mellitus (T2DM), regulation of energy metabolism and adiposity. Literature data indicate the action of TUDCA as chaperone molecule mechanism against endoplasmic reticulum stress (ER) that cause cell damage contributing to the pathophysiology of obesity and T2DM. Most studies attempted to demonstrate the action of TUDCA on insulin target tissues (liver, hypothalamus, brown adipose tissue) but the action of this compound on pancreatic endocrine morphofunction is not known, although different studies showed that bile acid can activate kinases such as protein kinase (PK) and C-mitogen activated kinase (MAPK) and regulate movement of Ca2 +. Thus, this project aims to characterize the effect of TUDCA and its mechanism of action on the secretion of insulin and glucagon in pancreatic islets isolated from mice. Also, check glucose homeostasis, pancreatic and morphofunction action of glucagon in ob / ob mice treated with TUDCA. (AU)
