Effects of physical exercise and low-level laser therapy on knee osteoarthritis induced by transection of the anterior cruciate ligament in rats

Osteoarthritis (OA) is a common cause of functional limitation and its prevalence has increased in recent decades. Thus, the development of therapeutic approaches that focus on avoiding damage and preventing its evolution can be helpful for rehabilitation. The therapeutic finding of moderate exercise and low-level laser therapy (LLLT) in the clinic treatment of OA is known, however, the signaling pathways activated by these resources remain unclear. The aim of this study is evaluate the effects of moderate exercise, LLLT and its association on the joint cartilage of rats with osteoarthritis induced by transection of the anterior cruciate ligament. Fifty male rats were divided into five groups (n=10): control; OA; OA plus exercise; OA plus LLLT; OA plus exercise plus LLLT. The treatments will be performed 2 weeks after surgery. The trained groups were submitted to an 8-week training program on a treadmill for 3 days/week (50 min/day, velocity 16 m/min). Will be used a GaAlAs diode laser (» =808 nm; P=30 mW; t=47 s; D=10 J/cm2; E=1.4 J; irradiance =3.8 mW/cm2) in two points of both lower limbs (medial and lateral joint line), immediately after each training session, 3 times for week. To evaluation the treatment effects will be performed cartilage morphological and morphometric analysis (cellularity, thickness, proteoglycan and collagen), lipid perodixation (TBARs), NO production (Griess reaction) and cytokines measurements, protein expression of IL-1², TNF-±, MMP-13, caspase-3 e SOD. These experimental may contribute to understanding of cellular systems and molecular mechanisms trigged by moderate aerobic exercise, LLLT and both in OA, optimizing the safe and effective rehabilitation of degenerative disease.
