Curcumin photodynamic effect in the treatment of induced periodontal disease: histological and immunohistochemical analysis

Abstract

The aim of this study is to evaluate by histologic and immunohistochemical analysis the effect of curcumin as a photosensitizer and Light Emitting Diode (LED) for antimicrobial photodynamic therapy (aPDT) as a periodontal treatment in induced periodontitis in rats. Periodontal disease will be induced at the first mandibular molar of 120 rats. After 7 days, the ligature will be removed. All animals will be divided according to the following treatments: G1 (n=24) - no treatment; G2 (n=24) - irrigation with curcumin (40µM); G3 (n=24) - irradiation with Light Emitting Diode (LED); G4 (n=24) - irrigation with curcumin and irradiation with LED (aPDT); G5 (n=24) - irrigation with methylene blue (MB) and irradiation with Low Level Laser Therapy (LLLT). LED (465 to 485 nm) or LLLT (660nm)will be applied at each buccal aspect of the mandibular first molar in contact with the tissue for 60 seconds. Eight animals in each group will be euthanized at 7, 15, and 30 days. The sections of the furcation area will be histologically analyzed. The markers RANKL and OPG will be analyzed immunohistochemically in both the bone and periodontal ligament in the furcation area. The immunohistochemical marker TRAP will be assessed by counting TRAP-positive cells located in the boundary of the most coronal portion of the bone tissue of the furcation region. The immunohistochemical values will be statistically analyzed with a significance level of 5%.(AU)
