Effect of viral oncoproteins E6 and E7 of HPV-16 in T cell response in patients with oropharyngeal squamous cell carcinoma

The Human Papillomavirus (HPV) has been associated with oropharyngeal squamous cell carcinoma as a possible risk factor. The viral oncoproteins E6 and E7 are capable of inhibit the production of some cytokines and also begin the production of others. So, the cellular response becomes deficient to the infection. The aim of this study is to evaluate the influence of the viral oncoproteins E6 and E7 of HPV-16 in the CD4+ T helper and CD8+ cells response in patients with oropharyngeal squamous cell carcinoma. The 20 participants will be divided in 10 patients diagnosed with oropharyngeal squamous cell carcinoma and 10 volunteers without cancer that will be submitted to the collect of tissue and peripheral blood samples to map the HPV-16 using Real Time PCR. T cells are isolated from peripheral blood, cultured in culture medium and stimulated by peptides E6 (16-40; 61-85; 91-115) e E7 (31-55; 46-70; 76-98) with 95% purity in three different concentrations. The level of the cytokines TNF±, IFN³, IL-2, IL-4 and IL-10 will be measured by ELISA technique in plasma and the culture medium of T cells after 1, 6 and 24 hours of stimulus. It will also investigate the effect of viral oncoproteins on lymphocyte proliferation by immunohistochemical tecnique for the rate of bromodeoxyuridine (BrdU) incorporation. Data from cell culture assays, as well as differences in constitutive lymphocyte immune response and response to stimulation with the viral oncoproteins E6 and E7 of the participants will be subjected to analysis of variance (ANOVA) followed by Bonferroni correction.
