Tissue engineering is an interdisciplinary science based on the combination of scaffolds, cells and growth factors aiming at repairing lost or damage tissues. Tissue engineering strategies as therapy for the treatment of bone defects resulting from trauma, infections and pathologic processess are of major intereste in oral and maxillofacial and orthopedic fields. Some preliminar results showed that a porous scaffold derived from Biosilicate®, named Bio-sca, presented a high osteogenic potential making it an attractive biomaterial to be used in bone tissue engineering. Thus, the aims of this study are: 1) in vitro evaluation of mesenchymal stem cells (MSC) and preosteoblasts behavior cultured on Bio-sca discs, and 2) in vivo evaluation of the effect of the combination of MSC and preosteblasts with Bio-sca on rat calvarial bone defects repair. MSC and preosteoblasts will be characterized by the expression of surface markers (CD13, CD29, CD31, CD44, CD73, CD90, CD105 e CD166, CD14, CD34, CD45) assayed by flow cytometry and gene expression of a panel of osteoblast markers alkaline phosphatase (ALP), runt-related transcription factor 2 (RUNX2), osterix (OSX), type I collagen (COL I), bone morphogenic protein-2 (BMP-2), bone sialoprotein (BSP), osteopontin (OPN) and osteocalcin (OC) by quantitative PCR. Cells cultured on Bio-sca discs will be assayed for cell proliferation, ALP activity, matrix mineralization formation, Runx2 and ALP protein expressions and the gene expresion of the same panel above. In vivo evaluations will be carried out in rat calvarial bone defects treated with Bio-sca+MSC, Bio-sca+preosteoblasts, Bio-sca and non-treated ones and evaluated by microtomographic and histomorphometric analyses
